Mechanics of the F-actin cytoskeleton.

چکیده
Dynamic regulation of the filamentous actin (F-actin) cytoskeleton is critical to numerous physical cellular processes, including cell adhesion, migration and division. Each of these processes require precise regulation of cell shape and mechanical force generation which, to a large degree, is regulated by the dynamic mechanical behaviors of a diverse assortment of F-actin networks and bundles. In this review, we review the current understanding of the mechanics of F-actin networks and identify areas of further research needed to establish physical models. We first review our understanding of the mechanical behaviors of F-actin networks reconstituted in vitro, with a focus on the nonlinear mechanical response and behavior of "active" F-actin networks. We then explore the types of mechanical response measured of cytoskeletal F-actin networks and bundles formed in living cells and identify how these measurements correspond to those performed on reconstituted F-actin networks formed in vitro. Together, these approaches identify the challenges and opportunities in the study of living cytoskeletal matter.

Effect of Stress-Fiber Inclusion on the Local Stiffness of Cell Cytoskeleton Probed by AFM Indentation: Insights from a Discrete Network Model
In this paper, we analyze the effect of stress-fiber inclusion on the stiffness of an actin random network. To do this, use a discrete random network model to analyze the elastic response of this system in terms of apparent Young’s modulus. First, we showed that for a flat-ended cylindrical AFM indenter the total indentation force has a linear relation with the indentation depth and the indente...
متن کاملI-6: Role of Actin Cytoskeleton during Mouse Sperm Acrosomal Exocytosis
Background: Mammalian sperm must undergo a process termed capacitation to become competent to fertilize an egg. Capacitation renders the sperm competent by priming the cells to undergo a rapid exocytotic event called acrosomal exocytosis that is stimulated by the zona pellucida (ZP) of the egg or progesterone. Over the years, several biochemical events have been associated with the capacitation...
متن کاملAssembly Kinetics Determine the Architecture of α-actinin Crosslinked F-actin Networks
The actin cytoskeleton is organized into diverse meshworks and bundles that support many aspects of cell physiology. Understanding the self-assembly of these actin-based structures is essential for developing predictive models of cytoskeletal organization. Here we show that the competing kinetics of bundle formation with the onset of dynamic arrest arising from filament entanglements and crossl...
متن کاملEffects of basic calponin on the flexural mechanics and stability of F-actin.
The cellular actin cytoskeleton plays a central role in the ability of cells to properly sense, propagate, and respond to external stresses and other mechanical stimuli. Calponin, an actin-binding protein found both in muscle and non-muscle cells, has been implicated in actin cytoskeletal organization and regulation. In this work, we studied the mechanical and structural interaction of actin wi...
متن کاملModulation of cellular mechanics during osteogenic differentiation of human mesenchymal stem cells.
Recognition of the growing role of human mesenchymal stem cells (hMSC) in tissue engineering and regenerative medicine requires a thorough understanding of intracellular biochemical and biophysical processes that may direct the cell's commitment to a particular lineage. In this study, we characterized the distinct biomechanical properties of hMSCs, including the average Young's modulus determin...
